基于网络药理学的"金银花-板蓝根"药对作用机制研究

OBJECTIVE:To probe into the material bases of efficacy and mechanism of making prescriptions of"lonicerae japonica-radix isatidis" herb couple based on the network pharmacology .METHODS: Data of chemical compositions , action targets and related diseases of lonicerae japonica and radix isatidis were retrieved to construct"drug-composition-target-disease" network and conduct classified enrichment analysis of gene ontology ( GO ) and pathway enrichment analysis based on Kyoto encyclopedia of genes and genomes ( KEGG ) , so as to investigate the functional mechanism of "lonicerae japonica-radix isatidis" herb couple .RESULTS: The "drug-composition-target-disease" network contained 2 kinds of drugs , 92 kinds of active compositions , 665 related targets and 609 kinds of related diseases.Key target spots involved prostaglandin-endoperoxide synthase-2 and so on, key diseases included various malignant tumors and inflammations .GO enrichment analysis contained 1680 GO items, of which 1367 involved biological process , 235 involved molecular function and 78 involved cellular compositions .KEGG pathway enrichment analysis contained 137 pathways , mainly were pathways of various malignant tumors and cardiovascular diseases.CONCLUSIONS:This study has preliminarily verified the basic pharmacological actions and the mechanism of"lonicerae japonica-radix isatidis" herb couple , and laid foundations for further revealing the functional mechanism .%目的:基于网络药理学,探讨"金银花-板蓝根"药对的功效物质基础和配伍机制.方法:通过数据库检索获得金银花和板蓝根的化学成分,作用靶点及相关疾病等信息,构建"药物-成分-靶标-疾病"网络,进行基因本体(gene ontology,GO)分类富集分析,基于京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)的通路富集分析,研究"金银花-板蓝根"药对的作用机制.结果:"药物-成分-靶标-疾病"网络包含2个药物,92种活性成分,相应靶标665个,相关疾病609种.关键靶点涉及前列腺素内过氧化物合酶2等,关键疾病涉及多种恶性肿瘤和炎症等.GO富集分析包含GO条目1680个,其中生物过程相关条目1367个,分子功能相关条目235个,细胞组分相关条目78个.KEGG通路富集分析包含KEGG通路137条,主要涉及多种恶性肿瘤信号通路和多种心血管疾病相关的信号通路.结论:本研究结果初步验证了"金银花-板蓝根"药对的基本药理作用及其机制,并为进一步深入揭示其作用机制奠定了良好基础.
