AnalogsofdUTPandUTPthat contain a biotinmolecule covalently bound to the C-5position ofthe pyrimidinering through an allylamine linker arm have been synthesized.Thesebiotin-labeled nucleotides areefficientsubstrates forava-rietyofDNAandRNA polynerases invitro. Polynucleotides con-taininglowlevelsofbiotinsubstitution (50'moleculesorfewerperkilobase) have denaturation, reassociation, and hybridizationcharacteristics similar. to those ofunsubstituted controls. Biotin-labeledpolynucleotides, bothsingle anddoublestranded, arese-lectively and quantitatively retained on avidin-Sepharose, evenafterextensivewashingwith8Murea, 6Mguanidinehydrochlo-ride, or 99%formamide. In addition, biotin-labeled polynucleo- tides can be selectively immunoprecipitated in the presence ofantibiotin antibody and Staphylococcus aureus protein A.
