The (Holey) study of mitochondria in apoptosis.

This chapter discusses the study of mitochondria in apoptosis. Accumulating evidence has shown that mitochondria also play an integral role in the death of a cell by apoptosis. Because of the mitochondrial cytochrome c release and its regulation by Bcl-2 family members appears to be a common mitochondrial change during apoptosis, the chapter focuses on the methods used to study cytochrome c release. The chapter also discusses methods to examine other mitochondrial parameters that may relate to the apoptosis, including the identification of proteins released from the mitochondria concomitantly with cytochrome c . Many inducers of apoptosis have shown to induce the release of cytochrome c from the mitochondria of cultured cells. Radiation, topoisomerase inhibitors, protein synthesis inhibitors, and protein kinase inhibitors are all commonly used apoptotic stimuli that trigger cytochrome c release. None of these toxins appears directly to induce the release of cytochrome c ; rather they use signaling pathways that may involve Bax, Bid, or other proapoptotic Bcl-2 family members. Various toxins act directly on mitochondria to induce cytochrome c release. Some of these toxins induce a phenomenon known as permeability transition (PT) in isolated mitochondria.
