Reports on yeast and mouse Sir2 proteins as nicotinamide adenine dinucleotide-dependent histone deacetylases. Yeast Sir2 as heterochromatin component that silences transcription at silent mating loci, telomeres and ribosomal DNA; Sir2 mutations supporting the idea that deacetylase activity accounts for silencing and recombinant suppression in vivo.
