Localizing receptors at the microscopic level has become as routine as many common histochemical techniques. The main approach that has been used is autoradiography because of the availability of radiolabeled binding ligands which have been well characterized in biochemical and pharmacological studies. Several general reviews have appeared on both theoretical and technical issues for autoradiographic receptor mapping 1–4. This article deals with two of the most current technical problems which have caused confusion and inaccuracy, even in recent studies — developing an appropriate means for quantitation of autoradiograms, and dealing with varying regional tissue quenching of β-missions from tritium-labeled ligands.
