[14] Immunoprecipitation of nitrotyrosine-containing proteins

Few endogenously nitrated targets have been identified in human disease; most likely as a result of limited immunodetection following standard western analysis .It has been observed that tyrosine nitration of manganese superoxide dismutase (MnSOD) has been identified in chronically rejecting human renal allografts. To achieve this observation, an immunoprecipitation technique has been developed that—when used in conjunction with western analysis and micro sequencing strategies—permits identification of specific nitrotyrosine- containing target proteins. Nitrotyrosine immunoreactivity has been reported in several human pathological conditions, including atherosclerosis, reperfusion injury, and brain ischemia. Numerous other disease states using nonhuman models have been shown to involve nitrotyrosine formation. This chapter is meant to provide the necessary information to perform the nitrotyrosine immunoprecipitation (IP). The development of the nitrotyrosine IP has proven to be a reliable method for identification of tyrosine nitrated proteins in several clinical situations. Undoubtedly, identification of precise nitrated targets in human disease will lead to a better understanding of the molecular basis of specific pathophysiologic processes.
