Prolactin potentiates insulin-stimulated leptin expression and release from differentiated brown adipocytes

The pituitary hormone () exerts pleiotropic effects, which are mediated by a receptor () present in numerous cell types including adipocytes. Brown adipose tissue (BAT) expresses uncoupling proteins (), involved in thermogenesis, but also secretes , a key hormone involved in the control of body weight. To investigate effects on BAT, we used the T37i brown adipose cell line, and demonstrated that are expressed as a function of . Addition of leads to activation of the /and MAP kinase signaling pathways, demonstrating that are functional in these cells. Basal and -induced expression were not affected by . However, combined with significantly increases expression and release, indicating that potentiates the stimulatory effect of as revealed by the recruitment of substrates and the activation of phosphatidylinositol 3-kinase. To explore the in vivo physiological relevance of action in BAT, we showed that content was significantly increased in BAT of -null compared with wild-type , highlighting the involvement of in the process. This study provides the first evidence for a functional link between and energy balance via a cross-talk between and signaling pathways in brown adipocytes.
