Reconstituted rattail collagen used as substrate for tissue cultures on coverslips in Maximow slides and roller tubes.

Liquifaction of the plasma clot and subsequent retraction of both the original explant and the outgrowth zone is a major problem of prolonged in vitro maintenance of many young (embryonic and newborn) and neoplastic tissues. Ehrmann and Gey have recently described a method of preparing the walls of roller tubes with a slice of coating of reconstituted rat-tail collagen as a substrate for growing cells in tissue culture. In the 29 different cell strains studies they noted the absence of lysis of the collagen gel and a promotion of outgrowth, without the retractions usually seen on glass. The method has been adopted for use on coverslips incorporated into either a classic Maximow slide assembly or a roller tube. Its usefulness has proved gratifying for the study of tissues from the central nervous system of chicken embryos and newborn kittens and rats. That it may be equally advantageous for other tissues occasions this communication.
