GERMINATION OF INDIVIDUAL MICROSCLEROTIA OF VERTICILLIUM DAHLIAE PRODUCED IN LABORATORY

Effects of ambient relative humidity, melanin inhibitor, sub-freezing, nutrient and soil on germination of microsclerotia of Verticillium dahliae was investigated. Results showed that germination rate of microsclerotia stored at regime 1 of 25% RH and 22℃ was reduced sharply with storage period within 30 days, while the germination of microsclerotia stored at regime 2 of 40% RH and 22℃ exhibited very stable and almost 100% even after 180 days. Melanin inhibitor [Tricyclazole and 4-chloro-3Methyl-2-(3H)-Benzothiazolone], sub-freezing (-20℃), different media (PDA, V8 juice, carrot juice and tomato juice agar) for producing microsclerotia did not enhance and maintain germination of microsclerotia stored at 25% RH. Meanwhile, natural dry soil can maintain the germination stable but not increase the germination ability of the microsclerotia after a long-term storage.
