Influence of F10 gene depletion or over-expression on the invasiveness of choriocarcinoma cell line JEG-3

Objective To investigate the influence of F10 gene depletion or over-expression on the expression of invasionrelated proteinase in choriocarcinoma cell line JEG-3.Methods Stable F10 gene over-expression or F10gene-silenced JEG-3choriocarcinoma cell lines were established by cell transfection and RNA interference technology.Thirty SPF nude mice aged from 4to 5 weeks were randomly divided into F10over-expression group(by injecting of F10 gene overexpressed JEG-3cells),F10gene-silenced group(by injecting of F10gene-silenced JEG-3cells)and control group(by injecting of non-treated JEG-3cells),with 10 mice in each group.The mice were sacrificed and the subcutaneous tumor tissues were gained 5weeks after the JEG-3cell injection.Western blot and immunohistochemical method were used to detect the expressions of matrix metalloproteinase(MMP)-2,MMP-8,MMP-11,MMP-16,MMP-19,tissue inhibitors of metalloproteinase-1(TIMP-1),and plasminogen activator inhibitor-1(PAI-1)in subcutaneous tumor tissues in three groups.Results The immunohistochemical method revealed the expressions of MMP-2,MMP-8,MMP-11,MMP-16 and MMP-19 were the highest in F10over-expression group(0.332±0.037,0.473±0.089,0.441±0.031,0.470±0.040,0.548±0.062),followed by control group(0.209±0.021,0.314±0.108,0.207±0.025,0.400±0.057,0.466±0.038)and F10 silenced group(0.049±0.013,0.168±0.034,0.149±0.024,0.240±0.024,0.175±0.029)in turns(P0.05).The expressions of TIMP-1and PAI-1 were the lowest in F10over-expression group(0.191±0.027,0.130±0.020),followed by control group(0.249±0.036,0.284±0.020)and F10 silenced group(0.310±0.018,0.463±0.115)in turns(P0.05).Western blot showed significant differences in the above invasion-related proteinase indexes among three groups(P0.05).Conclusion F10 gene could up-regulate the expressions of MMP-2,MMP-8,MMP-11,MMP-16 and MMP-19,and down-regulate the expressions of TIMP-1and PAI-1,which is involved in the proliferation,invasion and metastasis of choriocarcinoma cell.
