Research on the Expression of Thyroperoxidase mRNA and the Protein Biomarks of Ret,Galectin-3 and Mucin-1 in Thyroid Nodular Lesions

Objective To study the expression of thyroperoxidase(TPO)mRNA in thyroid nodular lesions and to analyze its correlations with the expression of Ret,Galectin-3 and Mucin-1 proteins.Methods TPO mRNA was analyzed with reverse transciption-polymerase chain reaction(RT-PCR) technique in the nodular lesions from thyroidotomy.The integral of apex areas in TPO and β-actin electrophoresis pieces was determined so that ratio of TPO/β-actin could be calculated which represented relative level of abundance of thyroperoxidase mRNA.The expression of Ret,Galectin-3 and Mucin-1 in the same lesions was tested by means of immunohistochemical technique.Results The expressive rate of TPO mRNA in the malignant group was lower than that in the benign one(P0.05).Semiquantitative analyses of 43 patients which expressed TPO mRNA showed ratio of the TPO/β-actin in benign group was higher than that in malignant one(P0.01).There were significant difference between benign and malignant lesions in the expression of Ret,Galectin-3 and Mucin-1(P=0.000).The positive rate of malignant group was higher than that of benign one.The expression of TPO mRNA was negatively correlated with that of Ret,Gal-3 and MUC1(P0.01,P0.05),and the expression of Ret,Gal-3 and MUC1 was positively correlated(P0.01).Conclusion High expression rate of TPO is one of the markers indicating benign thyroid lesions,whereas the expression rate of Ret,Galectin-3 and Mucin-1 is upregulated in thyroid carcinomas.Combined tests of TPO,Ret,Galectin-3 and Mucin-1 may be useful in the differential diagnosis of benign and malignant thyroid tumors.
