MAC ELISA test for serodiagnosis of JE was evaluated by comparing with HI test in a group of 232 suspected JE patients sera collected in 1985. Both of the test used antigen obtained from infected suckling mouse brain by sucrose-acetone extraction method. To detect IgM by MAC ELISA test by anti human IgM coated plate react with IgM from patients sera and the reaction is known by colour developing after serial addition of JE viral antigen, antiflavivirus human IgG peroxidase conjugated and ortho-phenylene diamine substrate. The colour reaction is measured by reading optical density at 492 nm and calculated in unit value. The sample which give the unit value more than or equal to 100 will be significant positive. By HI test using microtiter technique modified Clarde and Casals method. Thirty-seven percent of sample were positive by MAC ELISA test white twenty-four percent were positive by HI test. In case of inconclusived case by HI test, the MAC ELISA test can give seventeen percent positive. From the results showed the detection of IgM by MAC ELISA test is more sensitive and more rapid than HI test because of the untreated sera and require only single serum. Key words: J.E.
