Synthetic chemistry of base-modified DNA has provided a powerful tool for studying DNA-related subjects, in particular for understanding DNA damage and repair. This chapter first introduces basic concepts of DNA synthetic chemistry, and then reviews various approaches to chemical synthesis of base-modified DNA. It also presents the commonly used methods for purification and verification, illustrates applications of the synthetic base-modified oligomers and discusses the potentials of the synthetic chemistry.
