Describes three different crystal structures of the extracellular ligand-binding region of mGluR1, a metabotropic glutamate receptor involved in excitatory synaptic transmission in the central nervous system. Implication of the structures that glutamate binding stabilizes both the active dimer and the closed protomer in dynamic equilibrium; Description of its role in the initial receptor activation, which can be applied generally to G-protein-coupled neurotransmitter receptors that process extra-cellular ligand-binding sites.
