The interaction of infectious ribonucleic acid with a mammalian cell line. II. Kinetics of the formation of infectious centers.

The kinetics of the interaction of Mengo RNA and L cells in a suspended cell system have been studied. The formation of infectious centers in this system is directly proportional to the concentration of RNA in the incubation mixture, and to the total number, rather than concentration, of cells. The rate at which infectious centers are produced is strongly temperature dependent. At 37°, 27°, and 17°, the rates are linear for 15, 40, and 60 minutes, respectively, after which there is a rapid decline in the number of infectious centers recovered. The effect of temperature is adequately described by the Arrhenius equation for "irreversible" first-order reactions. The temperature characteristic, μ, of the system was found to be 18,700 cal/mole for the temperature range 25°–40°, and 33,900 cal/mole for the range 6°–21°. Some of the implications of the data are discussed.
