Tableting of coated pellets - Multiple unit dosage forms

Tableting of coated pellets into multiple unit system represents a dosage form that has technological and therapeutical advantages over a single unit dosage form. Compression of coated pellets is a technologically demanding task, because the coating must preserve its functionality, especially in case of film coatings, that are used to modify the drug release. To effectively tablet coated pellets, different cushioning excipients can be used, that limit or completely prevent the damage to the coating. Type of coating used is also very important, because different polymers have different mechanical properties, which can be improved with the use of plasticizers. Properties of multiple unit tablets depend also on compression force, content of coated pellets in tableting blend, pellet core (porosity, hardness), size of pellets, thickness of the coating and presence of additional protective coating. Manufactured multiple unit tablets must comply with the same pharmacopoeial tests as single unit dosage forms, the most critical parameters being suitably high hardness and low friability. Tableting of coated pellets is a challenging task that requires a person skilled in the art of tableting.
