Well-defined saccharide-polyacrylamide (PAA) conjugates containing the oligosaccharides (OS) which carry biological information and which are devoid of any other serologically active components are important antigenic tools for both immunochemical studies and diagnostic purposes. A general approach to the preparation of OS-PAA conjugates co-polymerization of unsaturated ligands (synthetic allyl or acrylamidoalkyl glycosides) or reducing saccharides isolated from natural sources with acrylamide has been developed. An alternative approach to OS-PAA conjugates by nucleophilic displacement of nitrophenyloxy groups in poly-(4-nitrophenylacrylate) with aminoalkyl glycosides is also reviewed. To illustrate the first synthetic route, the preparation of OS-PAA conjugates of synthetic fragments of and O-antigens and capsular polysaccharides of and is discussed. The OS-PAA conjugates obtained by both methods are efficient as coating antigens in EIA assays and were used in immunochemical studies and diagnosis of infectious diseases (salmonellosis).
