The permeability of keratinized and nonkeratinized oral epithelium to horseradish peroxidase.

Horseradish peroxidase was applied to the surface of keratinized and nonkeratinized oral epithelium of monkey, rabbit, and rat; in separate experiments peroxidase was injected subepithelially or intravenously into these animals. Biopsies taken at 20–60 min after application were examined for peroxidase activity with the electron microscope. Topically applied peroxidase did not penetrate deeper than 1–3 cell layers in either keratinized or nonkeratinized epithelium. When introduced subepithelially, peroxidase was visible in the connective tissue and extended through the intercellular spaces of the epithelium, into the granular layer of the keratinized layer or the "intermediate" cell region of nonkeratinized epithelium. In both tissues the limit of penetration coincided with the appearance of "membrane coating granules' at the cell margins, suggesting that the barrier to horseradish peroxidase in oral epithelium may be the result of intercellular material secreted by these granules.
