Relationship between the expression of tumor suppressor gene PTEN and the metastasis of colorectal cancer

Objective To investigate the relationship between expression of PTEN and the invasion or metastasis of colorectal cancer.Methods Using northern blot and immuno histochemistry,47 cases of colorectal carcinoma specimens were examined.The protein expression of PTEN was detected in three colorectal cancer cell lines with different metastasic ability by western blot.The human PTEN cDNA containing eukaryotic expression vectors pcDNA3.0 and eukaryotic expression vectors pcDNA3.0 only was transferred into human colorectal cancer cell line LOVO to investigate the changes of invasion and adhesion ability of LOVO by Boyden chamber and a glass plane combined with Laminin.Results The average value of PTEN mRNA and the positive rate of PTEN protein positive staining,which was found in the plasma,in colorectal cancer with lymph node or liver metastasis was lower than that in the tumor tissue without lymph node or liver metastasis ( P 0.05).The content of PTEN protein in LOVO,which had a higher ability of mestastase,was lower than that in HT 29,LS 174T,which had a lower ability of mestastasis.The adhesive rate of LOVO and LOVO/pcDNA3.0 PTEN to laminin at 20 min was (18.6± 1.4) % and (13.9±0.5)% respectively ( P 0.05),at 120 min was (71.2± 2.5)% and (56.0± 1.6)% ( P 0.05) respectively.The average number of LOVO and LOVO/pcDNA3.0 PTEN,which could transit through Boyden chamber was 11.7±1.7 and 7.5±1.6 ( P 0.05).Conclusion The expression of PTEN may play an important role in the metastastic process of colorectal cancer.
