The anaerobic metabolism of mitomycin C by rat liver homogenates and cell fractions was described. A TPNH-dependent system was localized in the microsomal (37000 x ) fraction. A more complex system, requiring DPN, malate, ATP, and Mg, was fully active only when mitochondria, microsomes and supernatant were recombined. When homogenates were supplemented with a TPNH-generating system no requirement for ATP, Mgor nicotinamide was evident although ATP had a sparing action with low concentrations of TPN.
