Potassium binding and oxidative phosphorylation in mitochondria and mitochondrial fragments

Relationships between K binding and oxidative phosphorylation were observed in intact mitochondria and in mitochondrial fragments. In the case of the intact mitochondria, evidence that the 2 processes are linked derives primarily from consideration of a common dependence of these functions upon the relative intactness of the mitochondrial structure. Experiments were also carried out which employ a preparation of smaller particle size derived from mitochondria that have been disrupted with digitonin. These mitochondrial fragments, which catalyze phophorylation, also retain the capacity to bind K. Respiration via the cytochrome system preserved bound K and increased the rate of exchange with radiopotassium of the medium. These effects did not appear to be dependent upon the presence of adenosine triphosphate or its active synthesis, but were abolished by 2,4-dinitrophenol. The probability of an association between the K binding site and the enzymes responible for coupling oxidation to phosphorylation is discussed.: The vesicular stage was absent in the development of cutaneous lesions in sheep.
