1. 1. The goal of this investigation was to determine whether caries-conducive streptoccoci form polyphosphates. 2. 2. A new technique of double isotopic labelling of washed stationary phase cells was employed. Polyphosphates were identified using the criteria of extraction and chromatographic characterization of 32P-labelled anionic components free of 14C. 3. 3. The barium precipitability criterion for the presence of polyphosphates was satisfied; however, it was insufficiently stringent. Highly anionic 32P-labelled compounds, free fo 14C, were isolated chromatographically and shown to behave like known polyphosphates by anion-exchange chromatography. 4. 4. Streptococci have not previously been shown to form polyphosphates. 5. 5. No 32P was incorporated into phospholipid. 6. 6. There is considerable phosphate turnover in the nucleic acids of these non-growing cells, devoid of an exogenous nitrogen source.
