A lignin precursor specifically enriched by carbon-13, i. e. coniferin-[side chain-α-13C], was synthesized and administered to the cavities of a growing rice plant (Oryza sativa L.) stalk. The 13C-enriched lignin-carbohydrate complexes (LCC) were obtained metabolically within 30 days. The 13C-NMR spectrum of the 13C-enriched LCC indicated clearly that lignin could be linked to carbohydrates through benzyl ester bonds and α-ketal linkages. It was also proved that β-O-4, β-β, β-5 and β-1 were the main substructures of LCC. After the extraction of 13C-enriched LCC (called LCCR-[α-13C]), the unsoluble residue was also enzymatically degraded and analyzed. It was found out that the lignin was linked to the carbohydrate by benzyl ether bonds at α-position of the side chain of the phenylpropane unit.
