Transcriptionally inactive oocyte-type 5S RNA genes of Xenopus laevis are complexed with TFIIIA in vitro.

Anextract fromwholeoocytes ofXenopus laevis wasshowntotranscribe somatic-type 5SRNA genes -100-fold moreefficiently thanoocyte-type 5SRNAgenes. Thispreference wasatleast 10-fold greater thanthe preference seenuponmicroinjection of5SRNAgenes into oocyte nuclei oruponinvitro transcription inan oocyte nuclear extract. The- 100-fold transcriptional bias infavor ofthesomatic-type 5SRNAgenes observed invitro inthewholeoocyte extract wassimilar tothetranscriptional biasobserved indeveloping Xenopus embryos. We also showed that inthewholeoocyte extract, apromoter-binding protein required for5SRNA genetranscription, TFIHA,wasboundbothtotheactively transcribed somatic-type 5SRNAgenes andtothe largely inactive oocyte-type 5SRNA genes. Thesefindings suggest thatthemechanism forthedifferential expression of5SRNAgenes during Xenopus development doesnotinvolve differential binding ofTFIIIA to5S RNA genes.
