Polyphenol Complexation

Natural polyphenols ( syn vegetable tannins ) are complex higher plant secondary metabolites. According to earlier definitions they are water soluble, possess relative molecular masses in the range 500 to 3,000 and besides giving the usual phenolic reactions, they have the ability to precipitate some alkaloids, gelatin and other proteins from solution. These complexation reactions have importance and wide ranging practical applications - in the manufacture of leather, in foodstuffs and beverages, in herbal medicines and in chemical defence and pigmentation in plants. The principal factors which influence the reversible association of polyphenols with other substrates are :- ( i ) solubility, solvation and desolvation; ( ii ) molecular size and conformational flexibility; ( iii ) general salt and specific metal ion ( e.g. Ca 2+ , Al 3+ ) effects; ( iv ) 'hydrophobic effects'; ( v ) hydrogen bonding via phenolic groups; ( vi ) the presence of tertiary amide groups in the co-substrate. The evidence for some of these observations is outlined as are methods to inhibit or enhance polyphenol-protein precipitation.
