A method of detecting a nucleotide sequence of a nucleic acid in a sample. The method comprises the steps of: (i) contacting, under hybridizing conditions, to form a strand of the nucleotide sequence with a control nucleic acid single-stranded, wherein a plurality of rare earth chelates is covalently linked through a polymeric structure of non-nucleic acid, soluble in water, to a nucleotide sequence complementary to the sequence to be detected, for forming a double-stranded nucleic acid, one of the strands carries the nucleotide sequence to be detected and the other strand porrte the reference nucleotide sequence, and (ii) detecting the double-stranded nucleic acid containing formation witnessed by measuring the fluorescence of the chelate rare earths contained in said nucleic acid double-stranded. The plurality of rare earth chelates has at least one metal ion selected from the group of rare earth Eu3 +, Sm3 +, Tb3 + and Dy3 + as a rare earth chelate. Witnesses described above are also claimed.
